Popović was born on December 9, 1881 in Sjenica (Сјеница) in modern-day Serbia, back then the Ottoman Empire. At the age of 6 his family moved to Serbia, and he entered the military academy, graduating in 1901. He fought as a Chetnik (guerilla fighter) against the Ottomans in the 'Macedonian Struggle' of 1905.
In the First and Second Balkan Wars in 1912 and 1913, he was the commander of the Chetnik Volunteer Detachment, comprising of some 2,250 soldiers. He established himself as a fearless, courageous and devoted leader and warrior, who had the utmost respect and trust from his men.
In July 1914 the First World War broke out, and Duke Vuk's Chetnik detachment was called up and attached to the Serbian Danube Division.
Duke Vuk went on to fight in the Battle of Cer in August 1914 where he suffered minor wounds, and was badly wounded in the abdomen in the Battle of Drina in October 1914. He also fought in the bloody Battle of Kolubara in November 1914 and was afterwards awarded the Order of Karađorđe's Star, being promoted to Lieutenant Colonel in the process.

In late 1915 he withdrew through the Albanian mountains together with the rest of the Serbian Army. In July 1916 his Chetnik Volunteer Detachment was positioned on the newly formed Macedonian Front, and they participated in the bloody Monastir Offensive in September 1916.
It was during a Serbian charge on the Gruništa Hill near Kajmakčalan that Duke Vuk was killed in combat by Bulgarian opposition on November 29, 1916, at the age of 35. Over 150 Serbian soldiers of his unit were killed in the action, but the Gruništa Hill was captured.
He was buried in the Zejtinlik Serbian Cemetery on the Macedonian Front but was transferred to Belgrade in September 1923 where he received a ceremony and state honors. It was stated that "when Duke Vuk was buried, Serbia was buried".
In 1936 a monument to Duke Vuk was erected in Belgrade, which still stands today.
